Yes you can know the flow between two servers, iff you have control over both the MTA.
What we do now is : to check the mail flowbetween abc.com - xyz.com , we send a mail to an invalid account at abc.com from xyz.com every 15 minutes. If the mail bounces back correctly, the flow is okay. The problem here is, a person need to check every fifteen minutes if the bounced mail as arrived.
