There are already lot of people doing the same. They are doing there job in a perfect way too. Why don't you spend your time in developing/improving packages. That will be more useful IMHO.

I don't know how to do this with with gnu packages but I ve done this once using debian distribution.
Try the following link. This is based on knoppix and is quite easy to customise.
http://www.knoppix.net/docs/index.php/KnoppixRemasteringHowto
You can also look at www.morphix.org. Essentially knoppix with some additional features thrown in.
Also If you want to start from a minimal GNU/Linux installation and build on top of it then take a look at www.damnsmalllinux.org. This could be a good point to start with.
All the three above are based on Debian distribution.

Before you start you need to decide on: 1. The objective of your distro 2. The minimum size of a working install 3. The mode of usage (str8 off the CD OR off the HDD) 4. The package management (RPM/DEB/etc.) 5. The installer (depends on package mgmt. choice)
I hope you have a group of people supporting you in this, as designing a distro, developing the distro, testing it, and iterating-till-success, is a very time consuming, and intensive process. Good Luck!
Oh, and by the way, I almost forgot, ;) here is a link to guide you (and anyone else who is interested and thinks of searching the archive): http://www.tldp.org/HOWTO/HOWTO-INDEX/os.html#OSDIST
